 Chemical Synthesis Laboratories

 

These facilities include three laboratories (SQ1, SQ2, SQ3) dedicated to the synthesis of novel non-radioactive molecules to be applied in the development of molecular/nanosized tools for application in nuclear imaging (PET or SPECT) or radionuclide therapy, namely for cancer theranostics.
The research activities developed in these facilities are focused on the synthesis of organic compounds, and inorganic or organometallic complexes of d- and f- transition elements. Organic compounds include biologically active molecules and bifunctional chelators, with or without biologically active fragments, to prepare the stable d- and f- transition metal complexes.
All the synthesized compounds are characterized by multinuclear NMR spectroscopy, X-ray crystallography, mass spectrometry and HPLC.

 Solid Phase Peptide Synthesis Laboratory

 


This Laboratory is dedicated to the solid phase synthesis, purification (semi-preparative HPLC) and characterization of biologically active peptides, taking advantage of a CEM-Liberty microwave automated peptide synthesizer.

 Laboratory for Animal Studies

 


In this laboratory biodistribution studies of radioactive probes are carried out in healthy or tumor bearing mice/rats. Pharmacokinetics and metabolic studies are also performed. This facility also includes an establishment for housing and care of small rodents with control of temperature, humidity and light, approved by the National Authorities.

Synthesis of Radiocompounds Laboratories 

 


These laboratories are under negative pressure and are equipped with hot cells, glove boxes and fume hoodsthat allow for the safe handling of radioactive compounds.
These facilities are also provided with High Performance Liquid Chromatography Systems (HPLC) coupled to UV-vis and Gamma Detectors, a Thin Layer Chromatography Scanner and an Electrophoresis Apparatus dedicated to the characterization and quality control of the radioactive compounds,
This infrastructure is also equipped with radiation monitors, dose calibrators and gamma counters for safety control and measurement of radioactivity. These laboratories are regularly used for training on radiopharmacy (students from ESTeSL and resident physicians in Nuclear Medicine)

Laboratories for Cell Studies 

 


This facility includes a laboratory for Cell Culture and a laboratory for In Vitro Studies of radioactive compounds
The Cell Culture laboratory is equipped with CO2 incubators, biohazard cabinets, a low temperature freezer (-80ºC) and a refrigerated centrifuge with conventional rotors, and inverted and conventional optical microscopes. The laboratory for In Vitro Studies of radioactive compounds is equipped with a fume hood, instruments for radiation measurement and a microplate UV-vis spectrophotometer.
In the Cell Culture laboratory a small collection of primary and cancer cell lines of animal or human origin is used to evaluate non-radioactive and radioactive compounds. The cell studies include: cytotoxicity and radiotoxicity assays, uptake, internalization, externalization and subcellular distribution studies as well as immunoreactivity, receptor affinity and mechanistic studies.
